چکیده

We describe an immunoassay for human pregnancyspecific f31-glycoprotein by use of fluorescein-labeled antibodies to this protein. The technique is generally similar to that of a “two-site” immunoradiometric assay but has the advantage of stability of tracer and avoidance of radiation hazard. However, the procedure requires relatively large quantities of antibody. Furthermore, the within-assay coefficient of variation (11.8%) is greater than that for radioimmunoassay (6.5%). This results from imprecision of the end-point detection, and some inherent difficulties in optimizing a “two-site” immunometric system. Concentrations of endogenous material as measured by the fluorometric assay are 1.9-fold higher than those obtained with radioimmunoassay, probably because the former detects multiple forms of this heterogeneous material.

Incidence of pregnancy-specific beta-1-glycoprotein in sera of malignant tumor patients.

Pregnancy-specific beta-1-glycoprotein (SP1) is said to be found only in sera of pregnant females. In this paper, SP1 in the sera of malignant tumor patients was investigated by means of Ouchterlony's method and counter immunoelectrophoresis (CIEP). On the examination by Ouchterlony's method, all samples were negative, but by CIEP, 34 out of 90 samples (38 percent) showed positive reaction; 25 ...

Particle-counting immunoassay (PACIA) of pregnancy-specific beta 1-glycoprotein, a possible marker of various malignancies and Crohn's ileitis.

Pregnancy-specific beta 1-glycoprotein (SP1) was assayed by particle-counting immunoassay (PACIA) with a sensitivity of 1 microgram/L. In serum from 50 men, the SP1 concentration was less than 1 microgram/L, whereas three of the specimens from 46 nonpregnant women had values exceeding 1 microgram/L. In 29% of 950 consecutive patients' sera, SP1 concentrations exceeded 1 microgram/L--in sarcoma ...

Monoclonal antibodies in analysis of oncoplacental protein SP1 in vivo and in vitro.

Three monoclonal antibodies were developed to the placenta-specific glycoprotein SP1. The antibodies were used in the characterization of SP1 in placental tissue, pregnancy serum and urine, cancer serum, and cell culture. The three antibodies reacted similarly with purified SP1 of placental origin in radio- and enzyme immunoassay, and all three decorated the syncytiotrophoblast layer of placent...

Radioimmunoassayof “ Pregnancy - Specific ’ ‘ - 3 1 - Glycoprotein ( SP 1 )

A radioirnmunoassay for SP1 (“pregnancy-specific” beta1-glycoprotein) is described in which a sensitivity of 0.1 tg/L was achieved by use of an antiserum with Ka 1.7 X lOll L/mol and delayed addition of a 1251-SP1tracer (specific acty, 25 Ci/g) that was stable for use in the assay for at least six to seven weeks. We also report an additional anti-SP serum, from an early bleeding, that displayed...

Epigenetic upregulation of large-conductance Ca2+-activated K+ channel expression in uterine vascular adaptation to pregnancy.

Our previous study demonstrated that pregnancy increased large-conductance Ca(2+)-activated potassium channel β1 subunit (BKβ1) expression and large-conductance Ca(2+)-activated potassium channel activity in uterine arteries, which were abrogated by chronic hypoxia. The present study tested the hypothesis that promoter methylation/demethylation is a key mechanism in epigenetic reprogramming of ...
